Varubi chemo chemotherapy ivTesaro Inc. has responded to reports of Varubi anaphylaxis in a recent warning letter.

Varubi (rolapitant) is an anti-nausea medication typically prescribed to chemo patients to prevent delayed chemo-induced nausea and vomiting. Chemotherapy patients often are affected by nausea after treatment because chemotherapy is incredibly hard on the body. The drug has been immensely popular and made an estimated $134 million in sales for 2017.

The drug is usually given on the first day of chemotherapy to prevent nausea and vomiting and can be given in a pill or injection form. The pill form of the drug was licensed in 2015 by Tesaro Inc. and the injection form was approved in October 2017.

In November 2017, chemotherapy patients began reporting Varubi anaphylaxis shortly after receiving an IV injection. Most Varubi anaphylaxis reactions occurred within the first few minutes of receiving the IV injection. Symptoms of a Varubi anaphylaxis reaction include wheezing, difficulty breathing, swelling of the face or throat, hives, flushing, itching, abdominal cramping, abdominal pain, vomiting, back pain, chest pain, hypotension, or shock.

Allergic reactions occur when the body’s immune system overreacts to certain substances and produces antibodies against them even if they aren’t harmful. Anaphylaxis is a severe allergic reaction that can occur minutes or even seconds after being exposed to an allergic trigger. The most common anaphylaxis reactions are to foods, insect stings, and medications.

Anaphylaxis causes the immune system to flush the body with flood chemicals which can cause individuals to go into shock. The reaction can cause a sudden drop in blood pressure and swelling of airways which can block breathing. Signs and symptoms include a rapid, weak pulse, nausea, and vomiting.

Treatment for anaphylaxis generally requires an injection of epinephrine, which can stop the body from going into shock, and a trip to the emergency room for further treatment.

On Jan. 16, 2018, Tesaro Inc. issued a warning letter following reports of Varubi anaphylaxis in patients who received the IV injection. The letter warns medical providers of the chance of “anaphylaxis, anaphylactic shock and other serious hypersensitivity reactions”, and urges providers to be vigilant for such reactions and stop administration of Varubi immediately should an adverse reaction occur.

The letter also warns that patients with allergies to legumes or other related allergens are at an increased risk for Varubi anaphylaxis. Tesaro Inc. estimates that at least 7,000 doses of Varubi have been administered to patients in the United States.
